Being new to EE I am curious what is the best way to get friendly URLs? In particular I am eager to get rid of index.php from every URL.

Removing index.php can be accomplished through URL rewriting in .htaccess. You should check here for more information.
You should probably also take a look at the Page Module, as it provides the ability to define URI segments and more closely control the URL for pages.
There are other items that could be discussed here, but I’ll refrain as those seem better fitted to one of the Community Support forums.
