We receive several mailer-daemon failure notices each day, it appears from people either using fake email addresses or mis-typing their addresses in the comments form. Is there anyway, within EE, to turn this functionality off?

No, these are generated by your mail server and have nothing at all to do with ExpressionEngine. You could talk to your administrator about where bounce notifications are sent; they may b able to over-ride the reply function and have them sent to a general address or simply not sent at all.
That also may not be possible; but worth exploring. =)
Personally, I just make an email rule for most of them so that they get trashed upon receipt. Some sites I do need to see them though, so it’s really project dependent.
